520 _aAbstract: Background: Consumption of faecal contaminated foods and drinks is associated with gastro-enteritis and diarrhoea diseases. There is limited information of faecal contamination of freshly extracted sugarcane juices in Mwanza, Tanzania. This study aimed to investigate the proportion of faecal contaminated freshly extracted sugarcane juices marketed at Nyamagana district, Mwanza Tanzania. Methodology: In the laboratory, juices samples were inoculated directly on MacConkey agar quantitatively using 10µl loop, plates were incubated aerobically at 37’C for 20-24 hours. TPC of bacteria growth was performed and then lactose fermenting colonies resembling E. coli were sub-cultured to obtain pure growth for further identification. Then, antibiotics susceptibility testing and sub-cultured on MCA plates supplemented with cefotaxime 2µg/ml for screening of extended spectrum β-lactamase production and STEC agar for screening of shiga toxin producing E. coli followed. STATA version 13.0 was used for data analysis. Results: The majority of the samples were positive with significant growth of TPC above 104 CFU/ml, but majority showed growth of TPC≥105 CFU/ml. The study found the prevalence of faecal contamination 61.2%. (n=71). Conclusion: This study observed that freshly extracted sugarcane juices marketed in Nyamagana district, Mwanza Tanzania are faecal contaminated. On provision of proper knowledge on hygiene and sanitation helped to reduce faecal contamination significantly.
